EVALUACIÓN DEL PROMOTOR nprA PARA LA EXPRESIÓN DE INTERFERÓN ALFA 2b EN Bacillus thuringiensis

Resumen
Various heterologous proteins are produced industrially due to the benefits provided by recombinant DNA technology. Interferon a-2b is a biopharmaceutical that improves the immune system's response to infections. This protein has been produced in multiple biological systems with varied results. Bacillus thuringiensis has been shown to be an organism that allows efficient production processes; however, there is little information on B. thuringiensis expressing heterologous proteins. Several studies have reported the production of interferon a-2 by microorganisms of the Bacillus genus. Neutral protease A (NprA) is massively produced by B. thuringiensis during the stationary phase, and its expression is maintained until the cell death phase. Therefore, the promoter that regulates NprA expression could be a key element to increase protein production. In addition, high NprA production has been described during liquid fermentation with B. thuringiensis. The use of the nprA gene promoter to produce interferon a-2 is proposed; the nprA promoter is expected to produce more interferon a-2 than the cry promoter. Two plasmid constructs were made with the nprA and cry promoters for the expression of interferon a-2, and the plasmids were inserted, by electroporation, into the B. thuringiensis strain HD73.
